Description
Local application of calyculin A (phosphatase inhibitor) can stimulate motility initiation. Phase-contrast time-lapse video of a stationary keratocyte exposed to a local application of calyculin A. Red pseudocolor indicates the calyculin A flow from the micropipette (positioned in the bottom left corner). The keratocyte polarized and initiated motility away from the source of calyculin A. 50x real time (frames were collected at 5-s intervals and are displayed at 10 frames/s). Cells were imaged in culture media (Leibovitz's L-15 medium without phenol red supplemented with 14.2 mM Hepes, pH 7.4, 10% FBS, and 1% antibiotic-antimycotic).
