Description
Micrograph following trifluoperizine (TFP) treatment, which apparently does not inhibit endocytosis but does inhibit movement of vesicles away from the parasomal sacs, coated and preendosomal vesicles and early endosomes become strongly labeled with HRP but all remain within the cortex. TFP seems to interfere with membrane movements into the cell so that even the products of coated pits arising from early endosomes, the carrier vesicles, remain in the cortex. TEM taken on 10/9/87 by R. Allen with Zeiss 10A operating at 80kV. Neg. 12,000X. Bar = 0.5µm. Adapted with permission from Fig. 15 published in J. Cell Sci. 101:449-461, 1992. The negative was printed to paper and the image was scanned to Photoshop. This digitized image is available for qualitative analysis. An unprocessed, high resolution version of this image (CIL:12615) is in the library and available for quantitative analysis. Standard glutaraldehyde fixation followed by osmium tetroxide, dehydrated in alcohol and embedded in an epoxy resin. Microtome sections prepared at approximately 75nm thickness.
